چکیده

Lipopolysaccharide induces a synergistic uptake of tritiated thymidine in peripheral blood lymphocytes (PBL) when cultured with phytohemagglutinin or concanavalin A as compared to PBL incubated only with phytohemagglutinin or concanavalin A. In this study we investigated which subpopulations(s) of PBL is involved in this synergistic increase in deoxyribonucleic acid synthesis. Separation of PBL into sheep erythrocyte rosette-forming cells (T cells) and non-rosette-forming cells (B cells) showed that the T cells were responsible for the increased uptake of radiolabel. PBL and T cells had similar dose-response profiles and kinetic patterns. Paralleling this augmented tritiated thymidine uptake was an increase in the number of cells undergoing blast transformation. Delayed-addition experiments showed that the two mitogens must be added within 12 h of each other for maximal augmentation to occur. Finally, preincubation of T cells with lipopolysaccharide had no demonstrable effect on the amount of concanavalin A uptake by these cells. This model may provide unique information about the activation of human peripheral blood T cells compared to activation of these cells by one mitogen.

Effect of amphotericin B and clotrimazole on lymphocyte stimulation.

Human lymphocytes were stimulated in vitro by phytohemagglutinin, concanavalin A, pokeweed mitogen, purified protein derivative-tuberculin, and allogenic cells. The deoxyribonucleic acid synthesis of the lymphocytes was inhibited in increasing degree by 4 to 8 mug of amphotericin B per ml of culture irrespective of lymphocyte stimulant used. The effect of clotrimazole on the deoxyribonucleic ac...

The Influence of Iron Loading and Iron Chelation on the Proliferation and Telomerase Activity of Human Peripheral Blood Mononuclear Cells

Background: Iron is an essential trace element in cell proliferation. Several investigations demonstrate that iron deprivation inhibits cell proliferation. However, the impact of iron on telomerase activity of activated lymphocytes remains unexplained to date. Objective: In this study, the effect of iron on the proliferation and telomerase activity of lymphocytes stimulated by phytohemagglutini...

Effects of Gamma Irradiation on Proliferation and IL-5 Production of Peripheral Blood Lymphocytes

Gamma irradiation is routinely used for suppression of lymphocyte function in transfusion and transplantation procedures. In recent years, some investigators focused on the effects of ionizing radiation on special aspects of lymphocyte function and considered the possibility of its clinical application for treatment of some immunological disorders. In this study, we evaluated the effects of fiv...

Different effects of concanavalin A and E-phytohemagglutinin upon lymphocyte glycosyltransferase activities.

We have studied glycosyltransferase activities in human lymphocytes stimulated with the plant mitogens E-phytohemagglutinin or concanavalin A (Con A) and have compared the results with activities found in resting lymphocytes. Compared to resting lymphocytes, Con A-stimulated lymphocytes possess an enhanced capacity to transfer the sugars, sialic acid, galactose, and N-acetyl-glucosamine from th...

Invesitgating the Radioprotective Effect of Brewed Rosa damascene in Comparison with Vitamin E on X-ray Induced Chromosomal Aberrations in Human Peripheral Blood Lymphocytes Using Micronucleus Assay in Binucleated Cells

Aims: More people, for occupational and therapeutic purposes, is exposed to its harmful effects including chromosomal abnormalities every day. Radioprotective agents are able to reduce these kinds of damages through mechanisms such as free radical scavenging, donating hydrogen to damaged molecules and increasing activity of antioxidant enzymes. Medicinal plants which have been traditionally use...
